  • ObjectiveTo investigate the role of expression and changes of mesencephalic astrocyte-derived neurotrophic factor (MANF) and nuclear factor (NF)-κB p65 in cerebral cortex of rats with cerebral ischemia-reperfusion injury (CIRI).MethodsThirty adult male SD rats were divided into Sham group and cerebral ischemia-reperfusion group (I/R group) according to random number table, 15 rats in each group. The model of focal CIRI in rats was established by modified thread embolization methods. In the Sham group, the thread embolization was not inserted into the internal carotid artery, and the other treatments were the same as the I/R group.Behavioral changes were observed by neurological deficit score 24 h reperfusion after 2 h ischemia, cerebral infarction area was observed by cranial magnetic resonance (MRI) and TTC staining, and cerebral cortex histopathological changes and cell damage in ischemic area were observed by HE staining. The relative protein levels of MANF and NF-κB p65 were detected by Western blotting, and the protein expression levels of NF-κB p65, CHOP and MANF were detected by immunohistochemistry. The subcellular localization of MANF and NF-κB p65 were observed by immunofluorescence staining. The apoptosis of cerebral cortex cells in the ischemic area was observed by TUNEL method, and the levels of TNF-α, IL-1β and IL-6 in serum of rats were detected by ELISA.ResultsCompared with the Sham group, the rats in the I/R group sh...

  • AbstractOxidative stress is considered to be one of the main causes of neural damage after injury. However, little is known about the changes in mRNA expression of antioxidant molecules that occur after injury and regeneration of the peripheral nerve. In the present study, the rat median nerve was transected, and transcriptional changes were studied at day 6 and day 12 after injury in both the proximal and the distal stumps, in the absence or presence of microsurgical repair. The expression profiles of the…

  • Background

According to the Classification and Catalogue of Occupational Diseases, brucellosis is one of the notifiable occupational infectious diseases, which occurs from time to time in the occupational population.

    Objective

To compare the work-related injury appraisal process and results of 13 cases of brucellosis at both provincial and municipal levels, analyze and summarize the bias in the practical work of labor capacity identification for occupational diseases such as brucellosis by appraisal management departments and experts, and propose suggestions for optimizing appraisal work.

    Methods

A comparative study was conducted on the objective examination results and labor capacity appraisal conclusions based on the occupational contact history, clinical diagnosis, occupational disease diagnosis staging, and labor capacity appraisal of 13 patients with brucellosis. The reasons for the inconsistency between the initial appraisal conclusion by institutions at the municipal level and the final appraisal conclusion by institutions at the provincial level were compared and analyzed.

    Results

All of the 13 patients with brucellosis applied for municipal-level labor capacity identification after being identified as work-related injuries, 11 of which did not receive a disability level, and 2 were rated as level 10 disability. Four of those who did n...

  •   Objective  To assess the effects of NOX2 (nicotinamide adenine dinucleotide phosphate oxidase) inhibitor gp91 ds-tat on lung injury of rats caused by gas explosion.  Methods  A total of 90 SD male rats were randomly divided into control group, gas explosion lung injury model group, and NOX2 inhibitor gp91 ds-tat group(n=30/group). The rats in the model group and gp91 ds-tat group were placed in the alleyway 240 m away from the point of detonation. After the explosion, the rats of the gp91 ds-tat group were immediately given once intraperitoneal injection of gp91 ds-tat (1 mg/kg). Then the rats were anaesthetized with blood taken from the abdominal aorta at 24 h, 72 h and 7 d (10 rats in each group) and then executed, respectively. ATP, α -KGDH, ICDHm and PFK were detected in frozen lung tissues. The expression levels of NOX2, CAT protein and mRNA as well as the expression of mitochondrial function -related protein genes were also detected with western blotting and real-time quantitative PCR in rat lung tissues.  Results  Lung bleeding and collapse were observed in the model group, and the expression of CAT, AMPK, TFAM decreased (P < 0.05), but the expression of NOX2 increased significantly(P < 0.05). Compared with the model group, levels of α-KGDH, ICDHm, PFK and ATP in gp91 ds-tat treatment group increased (P < 0.05), the relative mRNA expression of CAT...

  • Cardiac ischemia–reperfusion (I/R) injury results mainly from mitochondrial dysfunction and cardiomyocyte death. Mitophagy sustains mitochondrial function and exerts a pro-survival effect on the reperfused heart tissue. Mammalian STE20-like kinase 1 (Mst1) regulates chronic cardiac metabolic damage and autophagic activity, but its role in acute cardiac I/R injury, especially its effect on mitophagy, is unknown. The aim of this study is to explore whether Mst1 is involved in reperfusion-mediated cardiomyocyte death via modulation of FUN14 domain containing 1 (FUNDC1)-related mitophagy. Our data indicated that Mst1 was markedly increased in reperfused hearts. However, genetic ablation of Mst1 in Mst1-knockout (Mst1-KO) mice significantly reduced the expansion of the cardiac infarction area, maintained myocardial function and abolished I/R-mediated cardiomyocyte death. At the molecular level, upregulation of Mst1 promoted ROS production, reduced mitochondrial membrane potential, facilitated the leakage of mitochondrial pro-apoptotic factors into the nucleus, and activated the caspase-9-related apoptotic pathway in reperfused cardiomyocytes. Mechanistically, Mst1 activation repressed FUNDC1 expression and consequently inhibited mitophagy. However, deletion of Mst1 was able to reverse FUNDC1 expression and thus re-activate protective mitophagy, effectively sustaining mitochondrial homeostasis and blocking mitochondrial apoptosis in reperfused cardiomyocytes. Finally, we demonstrat...
